VS2010 Assembly Load Error

I am getting the following error when I try to build an ASP.NET 4 project in Visual Studio 2010: "Could not load file or assembly 'file:///C:Devproject runkinElmah.dll' or one of its dependencies. Operation is not supported. (Exception from HRESULT: 0x80131515)". I have verified that the dll does, in fact, exist, and is getting copied to the bin folder correctly. I have also tried removing and then re-adding the reference to the project.
The build only fails when I switch the Solution Configuration to "Release". It does not fail when the Solution Configuration is set to "Debug". The only difference between the two configurations (that I know of) is shown in the following Web.config transform, Web.Release.config:

I have tried using Fusion Log Viewer to track down the assembly binding issue, but it looks like it is finding and loading the assembly correctly. Here is the log:

I have created an application which runs fine in VS2010.Now i am trying to host that application using inetmgr.When i try to browse the page from inetmgr content view then i get the "Parser Error Message: Could not load file or assembly 'System.Web, Version=4.0.0.0, Culture=neutral, PublicKeyToken=b03f5f7f11d50a3a' or one of its dependencies. I have a VS2010 Web Application that uses the AjaxControlToolkit. I am able to build the Debug configuration. The location of the file is correct, and I just downloaded and unzipped there the Ajax CT. I have always built this app correctly before upgrading to VS2010 and ACT May 2010.

I've got an ASP.NET website that has abotu 10 projects makign it up which are compiled into DLLS and in the bin directory. ReportingContents is one of these projects. In the past I have had success in copying over a new version of the files (dll, pdb and xml) to the bin directory and recycled the app pool and it just picks up the new version on its own. This time I thought "I shouldn't have a debug version on the site" so I built as release and just copied up the dll and xml files and asked our deployment people to remove the pdb.When I was told it was all done I tried to access the site and got the above error message.The thing that surprises me is that the assembly it mentions is "Delete ReportingContents" and I don't know where the "Delete " might have come from in there.

I just migrated myDocuments folder to a new laptop (windows 7, 64 bit). The visual studio project WAS working on my desktop computer (windows XP pro).

I've add MSChart to a webpage and I'm also using a Survey page I got from 4GuysFromRolla (http://www.4guysfromrolla.com/articles/061604-1.aspx). Both work on my development machine but when I move them to my hosting site there are issues.
With MSChart page the chart is not displayed although it's included the Page Source code:

I installed a fresh machine with Win7 x64 Ult. wMS updates including 1/5/09, then installed VS2010 Ult beta with .Net V4.0. When I create a new web site, add the ajax references and controls, and use an ajax control on a generic aspx page then build I get the following popup: Find Source: ScriptObjectBuilder.cs and Find Source: ScriptControlBace.cs I downloaded the Ajax source file and point this to the file its requesting it doesn't like it. Anyone know what is going on with Ajax and VS2010?
